Why take it off? You will probably do more damage than good when you can’t see the roof unless you climb up on it.

Just a thought but, a clean roof makes it easier to see where you need to add sealant and where the sealant need to be replaced.

If a light spic and span brushing doesn’t work, I use a light bleach solution, and a paint roller on a broom handle to get rid of berry stains, mildew, etc from parking under trees. I do recommend keeping it clean so you can notice bad things before they get worse.

I also agree with Lynwood. Do nothing. Attempting to clean it will do more damage than leaving it there. If your normal roof cleaning products do not remove the stain, then "let it go!" It won't hurt anything and no one can see it but you when you get on the roof.
