The shortest route would be I-90 east to Wyoming Exit 9 at Ranchester.
Than 14 West to Cody.

About 10 miles out of Ranchester you will start climbing the mountain with numerous switchbacks and the like.

I said it's the "shortest way" but any other way will be miles out of your way.

I drove the reverse of your request last week towing my Montana. Not the shortest as Glenn provided, but certainly scenic. Here is the path I went (for your perspective):

From Rapid City I90 West to exit 58 (several large fuel stops off exit 58). Follow 16 West across Big Horn National Forest - we stayed in the forest at Boulder Park, a new national forest CG (no facilities, but very large back in sites). Big Horn has a couple of grades, but nothing that worries. We saw several moose while traveling through the pass. Stay on 16 through Worland (several fuel stops & grocery stores) and Greybull (both had many campgrounds). At Greybull take 14 West to Cody.

Take care,
Joe

I have been both 14 and 16 and I like 16 the best. If you go 14 make sure you do not turn on A14 or 14A whichever it is. It is known in the mountain book as OH MY GOD HILL, and it is a lulu. We drove it with the truck just to see it and I was glad I did not tow on it.
